You can watch, but play catch-up afterward, then watch it again so you'll get all the references and who's changed and how. ;)

Yay!! Goes and looks at this weeks synopsis *Are You There God? It's Me, Dean Winchester* well I think it should be fine. There will be some stuff that you might to need answers to but I think it should be alright.

WAIT. You'll have no context whatsoever for 4x01 if you skip everything else. I mean, you could watch 4x01, but it'd be so much better not to have some stuff from S1-3 spoiled.

I'd say, get caught up first... 4.01 won't make that much sense to you if you don't and I just generally prefer to watch things in order :)
