Season 3, No Rest For The Wicked. As Dean is getting closer to being dragged to hell, he's able to start seeing through the "veil" and can see the demons inside people. Well, near the beginning of the episode, he looks up at Sam and Sam's face goes all freaky. Why would it do that unless there was some kind of demon-y thing to make Dean see that? And yes, there was the demon blood from when he was a baby. But that was a teeny-tiny drop 25 years ago. I would think there needs to be something more there for this to happen.

And I point to two things to back that up: (1) Ruby in the S4 finale says that Dumbo didn't need the feather, that he had the powers inside him all along and (2) the psychic kids back in S2 - Eva, Jake, Andy, et al. - didn't need to drink demon blood to use their powers, they just had to let it happen (I believe Ava said it felt like flipping a switch).
So, I think that single drop from 25 years ago was enough. I think it wasn't the source of the power so much as the switch placed inside him.
I think what Dean was seeing as the veil was being lifted was all of Sam's as-yet-untapped demonic power, there, lurking beneath the surface, waiting to be switched on.
